Is the SL a designation of the black Penn aluminum spool and just the S the red aluminum spool?

There's a 500S, which is a plastic spool, a 500SM that is a metal spool, and 500SL that is aluminum spool. The majority of 500SL came with the red aluminum spool

 The 500S, & SM, & SL was Penn's attempt at using new material on the sideplates, called Lexan. It was also the only version of jigmaster with the take apart thumbscrew on the tailplate. Lexan only lasted a couple years and Penn went back to bakelite material and thumbscrew back on the headplate

Lexan sideplates tended to crack.
